How to Read the Evidence Tiers
Supplement evidence is a spectrum, and the honest way to communicate it is to define the tiers before assigning anything to them:

Caffeine has a half-life of roughly 5-6 hours, so a 300 mg dose at 5 p.m. leaves the equivalent of a cup of coffee in your blood at 11 p.m., and sleep is a bigger anabolic lever than any supplement. Stim Ceiling
Some "high-stim" pre-workouts stack 400 mg of caffeine with yohimbine, synephrine, and other stimulants. More is worse past your effective dose: anxiety, elevated heart rate, and wrecked sleep cost you more training quality than the extra stimulation adds. The FDA flags doses around 1,200 mg of caffeine as dangerous territory, and combining stimulants multiplies side effects faster than benefits.
Citrulline: The Pump Ingredient That Earned Its Spot
Citrulline raises blood arginine more effectively than taking arginine itself (it bypasses first-pass metabolism in the liver and gut), which increases nitric oxide production, vasodilation, and blood flow to working muscle. The research supports two forms: L-citrulline at 3-6g and citrulline malate at 6-8g. The classic Perez-Guisado and Jakeman trial found subjects performed significantly more bench press reps across multiple sets with 8g of citrulline malate and reported less soreness in the following 48 hours. Creatine monohydrate is the most proven supplement in existence for strength and lean mass. So why the asterisk? Timing is irrelevant. Creatine works by saturating your muscles over weeks, and pre-workouts that include it usually give you 1.5-3g instead of the 3-5g daily dose that saturation requires, only on the days you train.
Creatine in a pre-workout is a labeling convenience, and an underdosed one more often than not. Dietary Nitrates: The Underrated One
Beetroot-derived nitrate is the Tier 1 ingredient most pre-workouts skip. Nitrate converts to nitric oxide through a separate pathway from citrulline, and the McMahon meta-analysis found improvements in endurance performance, with effects strongest in efforts lasting a few minutes to about half an hour. It matters more for conditioning work than a five-rep squat set, and the effective dose (roughly 300-600 mg of nitrate, about 6.4-12.8 mmol) usually comes from concentrated beet juice shots rather than powder blends, where "beetroot extract 500 mg" is fairy dust compared with the doses that produced the research results.
Tier 2: Moderate or Mixed Evidence
Betaine: The Hype-Then-Correction Story
Betaine (trimethylglycine) is a textbook example of how supplement evidence evolves. The human data amounts to a handful of small trials: Bellar's 2015 study found 600 mg daily for six days improved isometric mid-thigh pull force versus placebo in a sample of 13 people. Taurine: Cheap, Safe, Modestly Supported
Taurine is in half the energy drinks on earth, and it has more legitimate data than its reputation suggests. Waldron's 2018 meta-analysis found a small but significant improvement in endurance performance at 1-6g doses. There is also evidence for reduced muscle soreness and a role in calcium handling within muscle cells. The effect is modest and most relevant to endurance and mixed conditioning work. At 1-2g it is a reasonable, inexpensive inclusion with a good safety record. Nobody should buy a pre-workout because of taurine, and nobody should avoid one because of it.
L-Tyrosine: A Stress Ingredient, Not a Strength Ingredient
Tyrosine is a precursor to dopamine and norepinephrine, and the human evidence shows it helps preserve cognitive performance under acute stress: sleep deprivation, cold exposure, sustained demanding tasks. Direct evidence that it improves lifting performance is essentially absent. In a pre-workout it may sharpen subjective focus, particularly stacked with caffeine, at doses around 500 mg-2g. Harmless, cheap, plausibly useful for the feel of a session, and without meaningful evidence for the barbell itself.
L-Theanine: The Caffeine Smoother
Theanine does one thing well: taken with caffeine (commonly at a 1:1 or 2:1 theanine-to-caffeine ratio, 100-200 mg), it takes the anxious edge off the stimulation while preserving alertness. The research base is about cognition and subjective calm rather than physical performance, which is why it sits in Tier 2. If pre-workout jitters ruin your focus, this single addition fixes more than another stimulant would.
Tier 3: Weak or No Evidence
Agmatine: A Mechanism in Search of a Human Trial
Agmatine sulfate is sold as a pump enhancer on the theory that it modulates nitric oxide synthase. The theory comes from cell and rodent work. Human trials demonstrating improved exercise performance, blood flow, or pumps do not exist in any meaningful form. Decades after it first appeared on labels, the performance evidence is still rodent-grade, and at the 500-1,000 mg commonly included, you are paying for a story. If a formula leans on agmatine for its "pump matrix" instead of a full citrulline dose, that tells you what the formulator was optimizing for, and it was the label.
Deer Antler Velvet: The IGF-1 Fantasy
Deer antler velvet is marketed as a natural source of IGF-1, a genuinely anabolic hormone, which makes for an irresistible story. The controlled evidence killed the story years ago: Sleivert's randomized trial found no meaningful differences in strength, endurance, or circulating hormones versus placebo. The IGF-1 in these products exists in trace amounts that would be digested like any other protein long before reaching your bloodstream. It briefly became famous through a 2013 sports scandal, and it survives on labels because the name sounds anabolic. Its presence on any label is a reliable signal to put the tub down.
BCAAs and Glutamine: Solved Problems
Branched-chain amino acids in a pre-workout duplicate what your protein intake already does, at premium prices and with an incomplete amino acid profile. Megadosed B-Vitamins and the Niacin Flush Trick
Many pre-workouts contain B12 at thousands of percent of the daily value. B-vitamins are involved in energy metabolism, so the label implies energy. Unless you are deficient, surplus B-vitamins do nothing acute, and the excess of the water-soluble ones leaves in your urine. Watch for niacin specifically: at 25-50 mg+ it produces a warm, flushed skin sensation that, like beta-alanine tingles, makes a product feel active without contributing anything to performance. Fancy Dust: Huperzine A, Yohimbine, and Friends
Huperzine A (an acetylcholinesterase inhibitor with essentially no exercise performance data), yohimbine (some fat-oxidation data, a real side-effect profile of anxiety and elevated heart rate), synephrine, and various tree barks and botanical extracts round out most labels. These exist at milligram doses to make a formula look sophisticated. None has convincing human strength or hypertrophy evidence. The pattern to notice: the less proven the ingredient, the more exotic the name, and the smaller the dose.

Proprietary Blend Red Flags
Everything above assumed you can read the doses. Proprietary blends exist to prevent exactly that. A blend lists a group of ingredients under a single total weight ("Pump & Power Matrix: 4,200 mg") with the individual amounts hidden as a "trade secret." Here is why that justification fails: effective doses are public knowledge. Every number in the table above came from published research anyone can read. There is no secret to protect. The only information a proprietary blend hides is how far below those numbers the product sits.
The arithmetic usually convicts the label on its own. Take a 4,200 mg blend listing citrulline malate, beta-alanine, betaine, agmatine, and tyrosine. Citrulline malate alone needs 6,000-8,000 mg. The entire blend weighs less than one properly dosed ingredient, and it claims to contain five. Ingredients are also listed in descending order by weight, so when caffeine (a few hundred milligrams) appears near the top of a multi-gram blend, everything after it is present in token amounts.
Red flags, in order of how fast they should end your interest:
- Any proprietary blend at all. Full-disclosure labels exist at every price point. Hiding doses is a choice, and there is only one reason to make it.
- Blend weight less than ~7g while claiming citrulline and beta-alanine. The math cannot work.
- Caffeine listed early in a blend. Everything below it weighs less than ~300 mg.
- A dozen or more ingredients in a modest scoop. Real doses take space; fifteen ingredients in 12g means most are dust.
- Feel-first ingredients doing the heavy lifting. Niacin flush plus beta-alanine tingles plus a big caffeine hit is the classic formula for making an underdosed product feel powerful in the first 20 minutes.
- Tier 3 marquee ingredients. Deer antler velvet or agmatine featured prominently tells you the formulator bet on marketing, not evidence.
The 30-Second Label Test
Check three numbers: caffeine (is it in your 3-6 mg/kg range?), citrulline (6-8g malate or 3-6g L-citrulline?), and whether every dose on the label is disclosed. A product that passes all three is probably honest. A product that fails the disclosure check fails everything, because you can no longer verify anything else.
Build Your Own for Half the Price
Once you know the doses, a branded tub is mostly a convenience fee. The evidence-backed core, bought as single-ingredient powders, costs roughly $0.50-0.70 per training day against the $1.50-2.50 per serving that mid-tier and premium tubs charge:
- Caffeine: 200 mg tablets cost a few cents each, or drink coffee, which arrives with its own evidence base. Dose to your 3-6 mg/kg range and your sleep schedule.
- Citrulline malate: 6-8g of bulk powder, taken 30-60 minutes pre-training. Sour; mix with anything flavored.
- Creatine monohydrate: 5g daily, any time of day, every day. Not actually a pre-workout ingredient, which is the point.
- Beta-alanine: 3.2g daily on its own schedule, split into two doses if the tingles bother you. Also not truly a pre-workout ingredient, also the point.
- Optional: 200 mg theanine alongside the caffeine if you run anxious; 1-2g taurine if your training leans conditioning-heavy.
Two of the four proven ingredients work through chronic saturation rather than acute timing, so the scoop-30-minutes-before ritual is doing less than the industry implies. What actually needs to be pre-workout is caffeine and citrulline. Everything else is a daily habit wearing a pre-workout costume. And no powder replaces the fundamentals it decorates: glycogen from adequate carbs, hydration with electrolytes if you sweat hard, and sleep.
My Read, Personally
At 38, my "pre-workout" is coffee dosed to about 3 mg/kg, 8g of citrulline malate in water, and that is the acute half. Creatine and beta-alanine run daily in the background, where the research says they belong. I stopped buying branded tubs after the third time I did the blend math in a store aisle and realized the tingle-and-flush combo was the product. Nothing in a proprietary matrix has out-trained that boring stack yet, and the boring stack costs less than half as much per session.
